Mounjaro provides several potential benefits when used appropriately as part of an individualized treatment plan.

• Helps Improve Blood Sugar Control- Tirzepatide helps regulate insulin and glucagon activity, which can reduce fasting and post-meal blood glucose levels.

• Once-Weekly Administration- Mounjaro is injected only once each week and may be administered at any time of day, with or without food.

• Helps Control Appetite- Its effects on GIP and GLP-1 receptors may increase fullness and reduce appetite, helping some patients better manage food intake.

• May Support Weight Reduction- Reduced appetite and food intake during tirzepatide treatment may lead to body-weight reduction, although individual results can vary.

• Recommended Maintenance Strength- The 5 mg once-weekly dose is a recommended maintenance dose and is commonly used after completion of the initial 2.5 mg treatment period.

• Convenient Multi-Dose KwikPen- Each Mounjaro 5 mg KwikPen contains four fixed weekly doses, providing approximately four weeks of treatment when used as directed.

Tirzepatide is a dual GIP and GLP-1 receptor agonist. These receptors play an important role in controlling blood glucose levels, insulin secretion, appetite and food intake. Mounjaro works through several complementary actions:

• It increases insulin release when blood glucose levels are elevated.

• It helps reduce inappropriate glucagon secretion.

• It slows gastric emptying.

• It can increase feelings of fullness.

• It helps reduce appetite and overall food intake.

• Over time, these effects can improve blood glucose control and may contribute to body-weight reduction.

Because tirzepatide increases insulin secretion in a glucose-dependent manner, the risk of hypoglycemia is generally greater when Mounjaro is combined with insulin or medicines such as sulfonylureas.

Mounjaro treatment is normally started with 2.5 mg injected under the skin once weekly for four weeks, after which the dose is commonly increased to 5 mg once weekly. The 5 mg strength is one of the recommended maintenance doses. If additional treatment effect is required and the medicine is well tolerated, the dose may be increased in 2.5 mg steps after at least four weeks on the current dose. Higher strengths include 7.5 mg, 10 mg, 12.5 mg and 15 mg once weekly, with 15 mg once weekly being the maximum recommended adult dose.

Patients should not increase, reduce or otherwise change their Mounjaro dosage without medical guidance. The most appropriate dose depends on treatment response, side effects, blood glucose control, other medicines being used and the individual's overall treatment plan.

Common Side Effects of Mounjaro 5 mg: Common side effects may include nausea, diarrhea, vomiting, constipation, reduced appetite, indigestion and abdominal discomfort. These effects are often mild to moderate and may improve as the body becomes accustomed to tirzepatide.

Serious Side Effects of Mounjaro 5 mg: Less common but serious reactions may include pancreatitis, severe allergic reactions, gallbladder problems, dehydration-related kidney injury and low blood sugar, particularly when Mounjaro is used with insulin or sulfonylureas. Seek medical attention if severe or unusual symptoms occur.

Mounjaro may not be appropriate for everyone. Patients should not use it, or should seek medical advice before treatment, if they:

• Are allergic to tirzepatide or any ingredient in the injection.

• Have previously experienced a serious allergic reaction to tirzepatide.

• Have a personal or family history of medullary thyroid carcinoma (MTC).

• Have Multiple Endocrine Neoplasia syndrome type 2 (MEN 2).

• Are pregnant or planning pregnancy.

• Have previously had pancreatitis.

• Have severe gastrointestinal disease or severe gastroparesis.

• Have diabetic retinopathy requiring monitoring.

• Are taking insulin or a sulfonylurea without discussing the risk of hypoglycemia with their healthcare provider.

• Are due to have surgery or another procedure requiring general anesthesia or deep sedation.

Mounjaro also delays gastric emptying, which should be considered before procedures requiring anesthesia or deep sedation.

Tirzepatide slows gastric emptying and can therefore affect the absorption of some medicines taken by mouth. Particular attention may be needed with:

• Insulin: may increase the risk of hypoglycemia.

• Sulfonylureas: may increase the risk of low blood glucose.

• Warfarin and other narrow-therapeutic-index medicines: closer monitoring may be required.

• Medicines dependent on rapid or predictable absorption: delayed gastric emptying may affect their absorption.

• Oral hormonal contraceptives: effectiveness may be reduced after starting Mounjaro or increasing the dose.

Patients using oral hormonal contraceptives should discuss switching to a non-oral method or adding a barrier method for 4 weeks after starting Mounjaro and for 4 weeks following each dose increase. Always tell the treating healthcare professional about prescription medicines, over-the-counter products, vitamins and supplements being used.

Unused KwikPen

• Store in a refrigerator at 2°C to 8°C (36°F to 46°F).

• Keep the pen in its original packaging to protect it from light.

• Do not freeze.

• Do not use the pen if it has been frozen.

• Keep it away from excessive heat and direct light.

After First Use

An in-use Mounjaro KwikPen may be stored in the refrigerator or at room temperature up to 30°C (86°F). The pen should be discarded after four weekly doses, 30 days after first use, or according to the applicable product instructions, even if medicine remains in the pen. Do not attempt to inject leftover medicine after the four intended doses have been administered.
