Mounjaro provides several potential benefits when used appropriately under medical supervision.

• Helps Improve Blood Sugar Control- Tirzepatide helps the body regulate insulin and glucagon responses, which can improve fasting and post-meal glucose control.

• Once-Weekly Administration- Mounjaro is injected only once each week, rather than every day. It may be taken at any time of day and does not need to be taken with food.

• May Support Weight Reduction- Weight reduction commonly occurs during tirzepatide treatment, although the amount differs substantially from person to person.

• Convenient Multi-Dose KwikPen - One 2.5 mg KwikPen contains four fixed weekly doses, providing approximately four weeks of treatment when used as directed.

Tirzepatide is a dual GIP and GLP-1 receptor agonist. These receptors are involved in regulating blood glucose, appetite and digestion. Activating them can help the body respond more appropriately to food intake and changing glucose levels. Mounjaro works through several complementary actions:

• It increases insulin release when blood glucose levels are elevated.

• It helps reduce inappropriate glucagon secretion.

• It slows gastric emptying, particularly after starting treatment.

• It can increase feelings of fullness.

• It can reduce appetite and overall food intake.

• Over time, these effects can help improve blood glucose control and may support body-weight reduction.

Because its insulin-related effects are largely glucose-dependent, tirzepatide by itself has a relatively low tendency to cause hypoglycemia. However, the risk increases when it is combined with medicines such as insulin or sulfonylureas.

The recommended starting dose of Mounjaro is 2.5 mg injected under the skin once every week. Dose increases should only be made under the direction of a healthcare professional and generally occur no sooner than after at least four weeks on the current dose.

Depending on the treatment plan and response, higher strengths may include:

5 mg → 7.5 mg → 10 mg → 12.5 mg → 15 mg once weekly

The maximum adult dosage listed in current U.S. prescribing information is 15 mg once weekly. Patients should not increase their dose on their own, even if they feel that the medicine is not working quickly enough.

• Common Side Effects of Mounjaro 2.5 mg - Common side effects of Mounjaro include nausea, diarrhea, vomiting, constipation, reduced appetite, indigestion and abdominal discomfort. These effects are often mild to moderate and may improve as the body adjusts to treatment.

• Serious Side Effects of Mounjaro 2.5 mg - Serious side effects are less common but may include pancreatitis, severe allergic reactions, gallbladder problems, dehydration-related kidney problems and low blood sugar, especially when used with insulin or sulfonylureas. Seek medical attention if severe or unusual symptoms occur.

Mounjaro may not be appropriate for everyone. Patients should not use it, or should obtain specialist medical advice before treatment, if they:

• Are allergic to tirzepatide or any ingredient in the injection.

• Have previously experienced a serious hypersensitivity reaction to tirzepatide.

• Have a personal or family history of medullary thyroid carcinoma where the applicable prescribing information lists this contraindication.

• Have Multiple Endocrine Neoplasia syndrome type 2.

• Are pregnant or planning pregnancy.

• Have previously had pancreatitis.

• Have severe gastrointestinal disease, particularly severe gastroparesis.

• Have diabetic retinopathy requiring monitoring.

• Are taking insulin or a sulfonylurea and have not discussed the increased risk of hypoglycemia.

• Are due to undergo surgery or a procedure requiring general anesthesia or deep sedation.

Mounjaro delays stomach emptying, and cases of pulmonary aspiration have been reported around procedures involving anesthesia or deep sedation. Patients should therefore inform their surgeon and anesthetist that they are using tirzepatide before a planned procedure.

Tirzepatide can slow stomach emptying and may affect how quickly certain oral medicines are absorbed.

Particular attention may be needed with:

• Insulin: increases the risk of hypoglycemia.

• Sulfonylureas: may increase the risk of low blood glucose.

• Warfarin and other narrow-therapeutic-index medicines: additional monitoring may be appropriate.

• Medicines requiring rapid absorption: delayed gastric emptying may affect their onset.

• Oral hormonal contraceptives: effectiveness may potentially be reduced after starting treatment or increasing the dose. 

Patients using oral hormonal contraception should discuss whether a non-oral contraceptive or additional barrier method is needed for 4 weeks after starting tirzepatide and for 4 weeks after each dose increase. 

Unused KwikPen

• Store in a refrigerator at 2°C to 8°C (36°F to 46°F).

• Keep it in the original packaging to protect it from light.

• Do not freeze.

• Do not use the pen if it has been frozen.

• Protect it from excessive heat and direct light. 

After First Use

The current U.S. KwikPen instructions allow an in-use pen to be stored refrigerated or at room temperature up to 30°C (86°F). Dispose of the pen:

• After four weekly doses,

• 30 days after first use,

• or according to the applicable product instructions,

Do not attempt to extract leftover liquid from the pen after the four intended doses have been used.
