Mounjaro can provide several potential benefits when used appropriately as part of an individualized treatment plan.

Tirzepatide is a dual GIP and GLP-1 receptor agonist. These receptors help regulate insulin production, glucagon secretion, appetite, food intake and digestion. Mounjaro works through several complementary actions:

• It increases insulin release when blood glucose levels are elevated.

• It helps reduce inappropriate glucagon secretion.

• It slows gastric emptying.

• It increases feelings of fullness.

• It may reduce hunger and overall food intake.

• These combined effects can improve blood glucose control and may contribute to body-weight reduction.

The risk of low blood sugar is higher when tirzepatide is used together with insulin or insulin-secretagogue medicines such as sulfonylureas.

Mounjaro treatment is generally started at a lower dose and increased gradually. A patient may be increased to 10 mg once weekly after using 7.5 mg once weekly for at least four weeks, when additional treatment effect is required and the current dose has been tolerated. Further increases may be made in 2.5 mg increments after at least four weeks on the current dose, with 15 mg once weekly being the maximum recommended adult dosage.

Patients should not move directly to the 10 mg dose or increase their dosage independently. Gradual dose escalation helps improve tolerability, particularly because gastrointestinal effects such as nausea, diarrhea or vomiting may become more noticeable after a dose increase.

Common Side Effects of Mounjaro 10 mg: Common side effects include nausea, diarrhea, reduced appetite, vomiting, constipation, indigestion and abdominal pain. These effects are usually gastrointestinal and may become more noticeable following a dose increase.

Serious Side Effects of Mounjaro 10 mg: Serious reactions may include pancreatitis, severe allergic reactions, gallbladder problems, dehydration-related kidney injury and hypoglycemia, particularly when Mounjaro is combined with insulin or sulfonylureas. Seek medical attention for severe or unusual symptoms.

Mounjaro may not be appropriate for everyone. Patients should not use it, or should obtain medical advice before treatment, if they:

• Are allergic to tirzepatide or any ingredient in the medicine.

• Have experienced a serious hypersensitivity reaction to tirzepatide.

• Have a personal or family history of medullary thyroid carcinoma (MTC).

• Have Multiple Endocrine Neoplasia syndrome type 2 (MEN 2).

• Are pregnant or planning pregnancy.

• Have previously experienced pancreatitis.

• Have severe gastroparesis or severe gastrointestinal disease.

• Have diabetic retinopathy requiring monitoring.

• Are using insulin or a sulfonylurea without discussing the risk of hypoglycemia.

• Are scheduled for surgery or a procedure involving general anesthesia or deep sedation.

Tirzepatide delays gastric emptying and may affect how quickly certain medicines taken by mouth are absorbed. Particular attention may be required with:

• Insulin: may increase the risk of hypoglycemia.

• Sulfonylureas: may increase the risk of low blood sugar.

• Warfarin and other medicines requiring careful monitoring: additional monitoring may be appropriate.

• Medicines requiring predictable oral absorption: delayed gastric emptying may affect absorption.

• Oral hormonal contraceptives: effectiveness may be reduced following treatment initiation or dose escalation.

Patients using oral hormonal contraceptives should discuss using a non-oral contraceptive method or adding a barrier method for 4 weeks following each Mounjaro dose increase. Always inform the healthcare professional about all prescription medicines, over-the-counter products, vitamins and supplements being used.

Unused KwikPen

• Store in a refrigerator at 2°C to 8°C (36°F to 46°F).

• Keep the pen in its original packaging to protect it from light.

• Do not freeze.

• Do not use if the pen has been frozen.

• Protect it from excessive heat and direct light.

After First Use

After first use, the Mounjaro KwikPen may be stored unrefrigerated at a temperature not above 30°C (86°F) for up to 30 days. The pen must then be discarded. Each KwikPen contains four intended doses, and any excess solution remaining after those doses have been administered should be discarded rather than used as an additional dose.
